Üçlü Negatif Meme Kanseri Hücre Hattında Androctonus crassicauda Ham Zehrinin Sitotoksik Aktivitesi, Apoptotik Etkileri ve DNA Etkileşim Çalışmaları

Özet

Yapılan çalışmalar birçok biyoaktif içeriğe sahip akrep zehrinin kanser tedavi etme potansiyeli olduğunu göstermiştir. Bu nedenle Androctonus crassicauda akrep zehrinin etkileri MDA-MB-231 üçlü negatif meme kanseri hücrelerine sitotoksik etkisi MTT yöntemiyle, apoptotik etkileri in vitro olarak belirlenmiştir. Mitokondriyal membran potansiyelindeki değişiklikler floresan boyama ile belirlenmiş ve apoptozla ilişkili genlerin (CASP3, CDKN1A, TP53, NOXA, PUMA, BCL2, BCL2L1 ve BAX) ifade düzeyleri RT-qPCR ile tespit edilmiştir. Bunun yanında, akrep zehrinin DNA'ya etki potansiyeli pBR322 plazmit DNA'sı kullanılarak incelenmiştir. Ham akrep zehrinin konsantrasyona bağlı olarak hücre canlılığını anlamlı ölçüde azaltığı gösterilmiştir (p<0,0001). Zehir uygulamasından 24 saat sonra mitokondriyal membran potansiyelinde önemli bir azalma gözlenmiştir. CASP3 geninin ekpresyon seviyesinde 4,7 kat azalış (p=0,0320), PUMA’da 12,5 kat artış (p=0,0238), CDKN1A 12 kat azalış (p= 0,0337) ve pro-apoptotik BAX geninde 2,33 kat anlamlı artış gözlemlenmiştir (p=0,0445). Ayrıca zehrin konsantrasyon bağımlı olarak DNA’da çift zincir kırıklarına neden olduğu belirlenmiştir.
Studies have shown that scorpion venom, which contains many bioactive compounds, has the potential to treat cancer. Therefore, the cytotoxic effects of Androctonus crassicauda scorpion venom on MDA-MB-231 triple-negative breast cancer cells were determined using the MTT method, and its apoptotic effects were determined in vitro. Changes in mitochondrial membrane potential were determined by fluorescent staining, and the expression levels of apoptosis-related genes (CASP3, CDKN1A, TP53, NOXA, PUMA, BCL2, BCL2L1, and BAX) were determined by RT-qPCR. In addition, the potential effect of scorpion venom on DNA was investigated using pBR322 plasmid DNA. Crude scorpion venom was shown to significantly reduce cell viability in a concentration-dependent manner (p<0.0001). A significant decrease in mitochondrial membrane potential was observed 24 hours after venom application. A 4.7-fold decrease in the expression level of the CASP3 gene (p=0.0320), a 12.5-fold increase in PUMA (p=0.0238), a 12-fold decrease in CDKN1A (p=0.0337), and a 2.33-fold significant increase in the pro-apoptotic BAX gene (p=0.0445) were observed. It was also determined that the venom causes concentration-dependent double-strand breaks in DNA.
