Background and Aim: The purpose of this study was to evaluate and compare the effects of different activation methods on the penetration of acid fuchsin-labelled sodium hypochlorite (NaOCl) at different concentrations (2.5% or 5.25%) and temperatures (room-temperature or 60°C) into the dentinal tubules via stereomicroscope. Materials and Methods: 128 mandibular premolar teeth were instrumented with ProTaper Universal System up to F3. The samples were randomly divided into 16 groups (n=8/ group) for final irrigation: Manual needle irrigation, EndoVac, EndoActivator or RinsEndo combined with 2.5%/5.25% NaOCl at room-temperature/60°C. Embedded samples into acrylic resin were sectioned transversely in 1mm thickness at 3-, 5- and 8-mm from the apex. Penetration depth and percentage were evaluated under a stereomicroscope. Data were analyzed statistically. Results: Regardless of temperature and concentration, RinsEndo resulted in statistically higher penetration depth and percentage values in all thirds (p<0.05). Higher temperatures also resulted in deeper penetration values (p<0.05). Conclusion: Activation of 60°C NaOCl at lower concentrations might be advantageous for improving its dentinal penetration instead of manual needle irrigation.
